The Id proteins (Inhibitors of DNA Binding/Differentiation) represent a family of dominant negative regulators of the basic helix-loop-helix transcription factors whose activities result in delayed cell differentiation and prolonged proliferation. The down-regulation of Id1 is necessary for differentiation to proceed in many cell lineages. Id1-deficient hematopoietic stem cells (HSCs) show increased cell cycling, but fail to efficiently self- renew. Id1 has been implicated in the maintenance of HSCs via its role in the cell cycle regulation.
