DetectX®Progesterone Metabolites ELISA试剂盒提供有孕酮标准品以生成测定的标准曲线,所有样品都从标准曲线中读出。将标准品或稀释的样品吸移到包被抗体的微孔板中以捕获兔抗体,将黄体酮-过氧化物酶结合物添加到孔中,通过向每个孔中的孕酮代谢物加入多克隆抗体来启动结合反应。孵育1小时后,洗涤板并加入底物。底物与结合的孕酮-过氧化物酶偶联物反应。短暂孵育后,停止反应,并在能够测量450 nm波长的微量滴定板阅读器中检测产生的颜色的强度。使用大多数酶标仪提供的软件对样品的稀释液进行适当校正后,可以计算出样品中孕酮的浓度。
Progesterone (P4, pregn-4-ene-3,20-dione) belongs to a class of hormones called progestogens, and is an essential regulator of female reproductive function in the uterus, ovary, mammary gland and brain. Progestogens are the primary hormones involved in the female menstrual cycle, gestation, and embryogenesis of humans and most other species. Progesterone can be metabolized and excreted as a variety of general progesterone by-products. Common metabolites include 5-reduced progesterone (pregnane), pregnanolones and hydroxyprogesterones. Measurement of progesterone metabolites provides vital data about reproductive status and is essential for studying reproductive and survival strategies of wildlife and endangered species. Likewise, assays measuring progesterone metabolites provide a clearer picture of hormonal regulation and reproduction.
